Results:
There are two types of each treatment of βeta-methylcyclodextrin (βMCD), fish oils and controls. Each treatment was ultrasonic homogenized at 0% and at 10% power to evaluate the optimal disruption of the plasma membrane. The first two images show controls at 0% and 10% homogenization. A majority of the proteins of caveolin and flotillin that are associated with lipid microdomains are contained in 1 - 4 fractions. In BMCD 0% and BMCD 10%, 0% and 10% homogenization of βMCD is shown. The majority of caveolin and flotillin is in fractions 1 – 4 for 0%. When βMCD is 10% ultrasonic homogenized, caveolin and flotillin associated with the lipid microdomain shift out of the first 4 fractions and into 6 fractions 5 - 10. A similar effect at 10% ultrasonic homogenization is demonstrated in the fish oil samples in the images labeled Fish Oil 0% and Fish Oil 10%.
